Western Blotting Enhanced Validation - Recombinant Antibody Technology Recombinant fragment of PDGF-C expressed in E. coli was probed with Cat. No. ZRB2443, Anti-PDGF-C, clone 4D14 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates PDGF-C (~25 and 12.5 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or PDGF-C peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB2443, Anti-PDGF-C, clone 4D14 ZooMAb® Rabbit Monoclonal. The binding of this antibody to PDGF-C peptide displayed a KD of 4.8 x 10-7 whereas binding to non-specific control peptide displayed a KD of 1.2 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Specificity: Clone 4D14 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Platelet-derived growth factor C (PDGF-C). It targets an epitope within 20 amino acids from the C-terminal half.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Platelet-derived growth factor C (UniProt: Q9NRA1; also known as PDGF-C, Fallotein, Spinal cord-derived growth factor, SCDGF, VEGF-E) is encoded by the PDGFC (also known as SCDGE, UNQ174/PRO200) gene (Gene ID: 56034) in human. PDGF-C is a disulfide-linked homodimeric growth factor that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to produce the mature form. It plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. It also plays an important role in wound healing and angiogenesis. It serves as a potent mitogen and chemoattractant for cells of mesenchymal origin. Its CUB domain is localized to amino acids 46 to 163. Proteolytic removal of the N-terminal CUB domain is essential for unmasking the receptor-binding epitopes of the core dom+D8ain. Cleavage after basic residues in the hinge region (region connecting the CUB and growth factor domains) gives rise to the receptor-binding form. PDGF-C expression is observed in the fallopian tube, vascular smooth muscle cells in kidney, breast and colon tissue, and in visceral smooth muscle of the gastrointestinal tract. It is highly expressed in retinal pigment epithelia. In the kidney, it is constitutively expressed in parietal epithelial cells of Bowman′s capsule, tubular epithelial cells, and in arterial endothelial cells. Higher levels of PDGF-C and have been reported in the malignant breast tumor microenvironment. In MCF-7 cells increased expression of PDGF-C is reported to enhance cell proliferation and tumor cell motility.
