Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from PANC-1 cells (Lane 1), Rat brain tissue (Lane 2), and Human brain tissue (Lane 3) were probed with Cat. Western Blotting Enhanced Validation - Recombinant Antibody Technology Recombinant bovine Connexin-36 was probed with Cat. No. ZRB2421, Anti-Connexin-36, clone 2D22 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Connexin-36 (~40 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of non-specific control peptide (Run 1) or Connexin-36 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB2421, Anti-Connexin-36, clone 2D22 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Connexin-36 peptide displayed a KD of 1.1 x 10-6 whereas binding to non-specific control peptide displayed a KD of 5.7 x 10-3.
ELISA Enhanced Validation - Recombinant Antibody Technology 50 ng of Connexin-36 peptide (Blue line) and a non-specific control peptide (Orange line) were analyzed by ELISA using various dilutions (starting at 3 g/mL) of Anti-Connexin-36, clone 2D22 ZooMAb® Rabbit Monoclonal. Alkaline phosphatase-conjugated Goat Anti-Rabbit secondary antibody was used for detection.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone 2D22 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Connexin 36. It targets an epitope within 17 amino acids from the C-terminal, cytoplasmic domain.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Gap junction delta-2 protein (UniProt: Q9UKL4; also known as Connexin-36, Cx36, Gap junction alpha-9 protein) is encoded by the GJD2 (also known as GJA9) gene (Gene ID: 57369) in human. Cx36 is a multi-pass membrane protein with 3 cytoplasmic domains, 4 transmembrane domains, and 2 extracellular domains. It is mainly expressed in neurons where it forms electrical synapses and in pancreatic β cells where it plays an important role in insulin secretion and glycemic control. It is regulated by intracellular pH and cytosolic magnesium ions, which can vary significantly under various physiological and pathological conditions. Its levels are upregulated under conditions of low intracellular pH and Mg2+. It exhibits a very low unitary conductance and low sensitivity to transjunctional voltage. Cx36 knockout (Cx36 -/-) mice exhibit impaired visual transmission and elevated auditory brainstem response threshold. Deletion of Cx36 in mice is shown to cause the loss of functional synapses between GABAergic interneurons in hippocampal slices.
