Immunocytochemistry Enhanced Validation - Recombinant Antibody Technology Immunofluorescent analysis of MCF-7 cell line was performed using a 1:300 dilution of Cat. No. ZRB1812, Anti-RARγ1, clone 1C22 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa FluorZooMAb® 488 (Green). Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.

Western Blotting Enhanced Validation - Recombinant Antibody Technology MCF-7 cell lysate was probed with Cat. No. ZRB1812, Anti- RARγ1, clone 1C22 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates RARγ1 (~55 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or RARγ1 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1812, Anti-RARγ1, clone 1C22 ZooMAb® Rabbit Monoclonal. The binding of this antibody to RARγ1 peptide displayed a KD of 1.1 x 10-6 whereas binding to non-specific control peptide displayed a KD of 2.9 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone 1C22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Retinoic Acid Receptor 1 (RAR 1). It targets an epitope within 16 amino acids from the N-terminal region.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Retinoic acid receptor gamma (UniProt: P13631; also known as RAR-gamma, Nuclear receptor subfamily 1 group B member 3) is encoded by the RARG (also known as NR1B3) gene (Gene ID: 5916) in human. RAR 1 is a member of the nuclear hormone receptor family that serves as a receptor for retinoic acid. Three isoforms of RAR have been described: RAR-a , RAR-β, and RAR- that are expressed in temporal- and tissue-specific patterns beginning early in development and continuing throughout life. RAR- shares about 90% homology with RAR-a and RARβ. It is composed of three domains: a modulating N-terminal domain, a DNA-binding domain (aa 90-155) and a C-terminal ligand-binding domain. It contains two NR C4-type Zinc finger domains (aa 90-110 and 126-150). Four different isoforms of RAR- have been described that are produced by alternative splicing and they differ only in their N-terminal regions. RARs form heterodimers with member of the RXR gene family that also has three different isomers of RXRs (RXR-a, RXR-β, RXR- ) and are expressed in distinct patterns. The binding of all-trans-retinoic acid to RAR in an RAR-RXR heterodimer serves to activate the transcription factor complex, which, when bound to specific retinoic acid response elements (RARE) in target genes can recruit additional protein factors that activate or repress gene expression. The activity of the RARs is further modulated by posttranslational modifications. In human epidermis, RAR- is reported to be the predominant RAR that accounts for about 87% of RAR protein and RAR-a makes up the remainder.
