Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from HeLa cells (Lane 1), HEK293 cells (Lane 2), and L6 cells (Lane 3) were probed with Cat. No. ZRB1771, Anti-Catalase, clone 1C21 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Catalase (~58 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or Catalase peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1771, Anti-Catalase, clone 1C21 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Catalase peptide displayed a KD of 6.9 x 10-7 whereas binding to non-specific control peptide displayed a KD of 2.0 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone 1C21 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Catalase. It targets an epitope within 17 amino acids from the C-terminal region.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Catalase (UniProt: ; also known as CAT, EC:1.11.1.6) is encoded by the CAT gene (Gene ID: 847) in human. Catalase is an antioxidant enzyme that is present in all aerobic organisms. It is predominantly localized in peroxisomes and the inner mitochondrial membrane. It catalyzes the conversion of hydrogen peroxide (H2O2) into water and oxygen and thereby protects cells from oxidative damage. Catalases can be divided into three classes: monofunctional catalase (typical catalase), catalase-peroxidase, and Mn-catalase. The first two groups are heme-containing enzymes, whereas the third group contains non-heme (manganese) catalase. Human catalase is reported to contain four identical subunits of 62 kDa and each subunit contains four distinct domains and one prosthetic heme group. The four domains include a N-terminal arm that contains a distal histidine that is reported to be important for catalase reaction; a β-barrel domain that has eight β-barrels arranged in an antiparallel fashion with six -helical insertions, creating a hydrophobic core of the protein; a connection domain that contains the tyrosine residue that binds the heme group; and an -helical domains, which is shown to be important for NADPH binding. During the enzymatic reaction that leads to H2O2 destruction, catalase is first oxidized to a hypervalent iron intermediate (compound 1), which is then reduced back to the resting state by a second H2O2 molecule.
