No. ZRB1742, Anti-NOVA2, clone 2J16 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Protein was visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates NOVA2 (~52 kDa).
Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from E16 Rat brain (Lane 1) and E16 Mouse brain E16 (Lane 2) tissues were probed with Cat. No. ZRB1742, Anti-NOVA2, clone 2J16 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates NOVA2 (~65 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or NOVA2 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1742, Anti-NOVA2, clone 2J16 ZooMAb® Rabbit Monoclonal. The binding of this antibody to NOVA2 peptide displayed a KD of 2.4 x 10-6 whereas binding to non-specific control peptide displayed a KD of 3.7 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Specificity: Clone 2J16 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects RNA-binding protein NOVA2. It targets an epitope within 20 amino acids from the N-terminal half.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: RNA-binding protein Nova-2 (UniProt: Q9UNW9; also known as Astrocytic NOVA1-like RNA-binding protein, Neuro-oncological ventral antigen 2) is encoded by the NOVA2 (also known as ANOVA, NOVA3) gene (Gene ID: 4858) in human. The neuron specific RNA-binding proteins NOVA1 and NOVA2 are highly homologous alternative splicing regulators. Both NOVA1 and NOVA2 have three K homology (KH) domains. They are described as autoantigens in the human autoimmune neurological disease known as paraneoplastic opsoclonus-myoclonus ataxia (POMA) that is characterized by loss of motor control in limbs and eyes. NOVA proteins regulate alternative splicing in neurons by binding pre-mRNA in a sequence-specific manner to activate exon inclusion or exclusion. NOVA2 binds RNA with high affinity and with sequence specificity that differs from NOVA1. Its expression is restricted o astrocytes. In close association with NOVA 1, it performs unique biological functions in different brain areas and cell types. NOVA2 binding to an exonic 5′-YCAY-3′ cluster changes the protein complexes assembled on pre-mRNA, blocking U1 snRNP binding and exon inclusion, whereas binding to an intronic 5′-YCAY-3′ cluster enhances spliceosome assembly and exon inclusion. It is reported to regulate alternative splicing events of several axon guidance related genes during cortical development. It controls RNA splicing in both inhibitory and excitatory neurons. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
