Western Blotting Enhanced Validation - Recombinant Antibody Technology HepG2 (Lane 1), HeLa (Lane 2), and NIH3T3 (Lane 3) cell lysates were probed with Cat. No. ZRB1738, Anti-LLGL2, clone 2H12 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates LLGL2 (~110 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or LLGL2 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1738, Anti-LLGL2, clone 2H12 ZooMAb® Rabbit Monoclonal. The binding of this antibody to LLGL2 peptide displayed a KD of 1.4 x 10-6 whereas binding to non-specific control peptide displayed a KD of 2.0 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone 2H12 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects LLGL2. It targets an epitope within 25 amino acids from the C-terminal region.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: LLGL scribble cell polarity complex component 2 (UniProt: Q6P1M3; also known as HGL, Lethal(2) giant larvae protein homolog 2) is encoded by the LLGL2 gene (Gene ID: 3993) in human. LLGL2, a scaffolding protein with 14 WD repeats, regulates the establishment of apical basal polarity in epithelial cells. It is a part of a complex consisting of GPSM2/LGN, aPKC/PRKC1, and PARD6B/Par-6, which ensures the correct organization and orientation of bipolar spindles for normal cell division. This complex plays roles in the initial phase of the establishment of epithelial cell polarity. It is localized in the perinuclear structure and faintly at the cell-cell contacts sites in the interphase. During metaphase it is localized at the cell periphery. LLGL2 undergoes phosphorylation at Ser 653 by PRKC1 and this phosphorylation is enhanced during cell polarization induced by calcium. Phosphorylation may occur during the cell-cell contact-induced cell polarization and may contribute to the segregation of LLGL2 from the PRKCI/aPKC and PARD6B/Par-6 complex. Overexpression of LLGL2 is shown to inhibit the tight junction formation. Overexpression of LLGL2 has also been reported in ER+ breast cancer where it promotes cell proliferation under conditions of nutrient stress. It regulates cell surface levels of leucine transporter, SLC7A5, by forming a trimeric complex with SLC7A5 and YKT6, to promote leucine uptake and cell proliferation. In mammals LLGL2 is shown to be essential for proper branching morphogenesis during placental development.
