Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of of non-specific control peptide (Run 1) or PLD2 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1629, Anti-PLD2, clone 6F13 ZooMAb® Rabbit Monoclonal. The binding of this antibody to PLD2 peptide displayed a KD value of 1.7 x 10-6 whereas its binding to non-specific peptide displayed a KD of 3.6 x 10-3.
Flow Cytometry Enhanced Validation - Recombinant Antibody Technology Staining of one million NIH3T3 cells was performed using 1 μg of Cat. No. ZRB1629, Anti-PLD2, clone 6F13 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-RabbitIgG secondary antibody.

Target description: Phospholipase D2 (UniProt: O14939; also known as EC:3.1.4.4, PLD 2, hPLD2, Choline phosphatase 2, PLD1C, Phosphatidylcholine-hydrolyzing phospholipase D2) is encoded by the PLD2 gene (Gene ID: 5338) in human. PLD2 is a peripheral membrane protein that displays ubiquitous expression and plays a role in cell membrane lipid reorganization and serves as a key cell signaling protein. It is activated by a wide variety of G-protein coupled receptors and receptor tyrosine kinases in response to a wide variety of agonists. It hydrolyzes the membrane phospholipid phosphatidylcholine to choline and the lipid messenger phosphatidic acid (PA). PA can act as a central lipid second messenger for various signaling pathways and participates in many physiological processes, including cell proliferation, migration, and cytoskeletal organization. PA is also involved in the activation of Ras‐GTPase by recruiting its exchange factor Sos or Rac1to the plasma membrane. PLD2 is reported to positively regulate proliferation of CD8+ T cells that play a critical role in cancer cell elimination and suppression of tumor progression. PLD2 is reported to degrade HIF-1 and thereby reduces VEGF secretion by endothelial cells that results in reduction of tumor neovascularization and growth. PLD2 has PX and PH domains at its regulatory N‐terminus that are putatively responsible for its GEF activity. It also has two conserved HKD domains are responsible for the lipase activity and conversion of PC into PA. Its catalytic region is localized within amino acids 441-788.
