Western Blotting Enhanced Validation - Recombinant Antibody Technology Recombinant IFN-g protein was probed with Cat. No. ZRB1620, Anti-IFN-g, clone 5K15 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates IFN-g (~15 kDa for recombinant IFN-g).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of non-specific GST protein (Run 1) or IFN-g protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1620, Anti-IFN-g, clone 5K15 ZooMAb® Rabbit Monoclonal. The binding of this antibody to IFN-g displayed a KD value of 2.3 x 10-8 whereas its binding to non-specific GST protein displayed a KD of 2.7 x 10-3.
Flow Cytometry Enhanced Validation - Recombinant Antibody Technology Staining of one million Human PBMC was performed using 1 μg of Cat. No. ZRB1620, Anti-IFNγ, clone 5K15 ZooMAb® Rabbit Monoclonal. Histogram represents the gated R3 population shown in the dot plot.

Target description: Interferon- g (UniProt: P01579; also known as IFN-gamma, IFN- g, Immune interferon) is encoded by the IFNG gene (Gene ID: 3458) in human. IFN-g is a pleiotropic molecule with antiproliferative, pro-apoptotic, and antitumor properties. It is a type II homodimeric interferon that is synthesized with a signal peptide (aa 1-23) and a propeptide (aa 162-166) that are subsequently cleaved off to produce the mature form. It is secreted predominantly by activated lymphocytes such as CD4 T helper type 1, CD8 cytotoxic T cells, g T cells, and natural killer cells, and antigen-presenting cells. Its expression is induced by mitogens and cytokines. IFN- g signaling coordinates several biological responses involved in host defense and immune surveillance, establishment of adaptive immunity, and the regulation of inflammation. Its diverse biological activities are initiated by IFN- g -mediated aggregation of at least two different cell surface receptors: IFN g R1 and IFN g R2. IFN-g is reported to activate macrophages toward a pro-inflammatory profile increasing their phagocytic ability and enhancing their microbial killing activity. It is also shown to upregulate cell surface MHC class II on antigen presenting cells, thus promoting peptide-specific activation of CD4 T cells. Mutations in IFNG gene are known to cause aplastic anemia that is characterized by failure of bone marrow to produce adequate number of peripheral blood elements leading to peripheral pancytopenia and hypoplasia. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
