Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from L6 (Lane 1), U251 (Lane 2), NIH3T3 (Lane 3), K562 (Lane 4), and HEK293 (Lane 5) cells were probed with Cat. No. ZRB1584, Anti-Vimentin, clone 2I13 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Vimentin (~54 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of non-specific control peptides (Run 1) Vimentin peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1584, Anti-Vimentin, clone 2I13 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Vimentin peptide displayed a KD value of 5.5 x 10-7 whereas its binding to non-specific control peptides a KD of 6.9 x 10-4.

Target description: Vimentin (UniProt: P08670) is encoded by the VIM gene (Gene ID: 7431) in human. Vimentins are class-III intermediate filaments found in various non-muscle cells. It is the major subunit protein of the intermediate filaments of mesenchymal cells. Vimentin is attached to the nucleus, endoplasmic reticulum, and mitochondria, either laterally or terminally. It is usually expressed by cells undergoing cell-to-cell adhesion, EMT, and cytoskeleton remodeling. It is also involved with the intracellular transport of proteins between the nucleus and plasma membrane. Vimentin is also involved in positioning autophagosomes, lysosomes and the Golgi complex within the cell. It has also been implicated to be involved in the rate of steroid synthesis via its role as a storage network for steroidogenic cholesterol containing lipid droplets. Vimentin consists of a central -helical rod domain. The N-terminal part of the rod domain is a coil 1 motif that is organized into sequences of seven amino acid residues, where every first and fourth residue is hydrophobic. The C-terminal half of the rod domain has a coil 2 motif that shows a hydrophobic pattern. The vimentin structure shown to be dynamic in cells and can be rearranged to form filaments or the soluble tetrameric protein. The head and tail domains of vimentin contain over 35 phosphorylation sites and phosphorylation of its serine residues is reported to inhibit its subunit polymerization, thus promoting the disassembly of vimentin filaments and increasing its solubility. These phosphorylations also regulate protein-protein interactions and intracellular signaling.
