Western Blotting Enhanced Validation-Recombinant Antibody Technology Recombinant CCPG1 (Left image) and A549 cell lysate (Right image) were probed with Cat. No. ZRB1582, Anti-CCPG1, clone 1F15 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates CCPG1 (~ 34 kDa for recombinant fragment of CCPG1; ~90 kDa in A549 cell lysate).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of non-specific GST protein (Run 1) or recombinant CCPG1 protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1582, Anti-CCPG1, clone 1F15 ZooMAb® Rabbit Monoclonal. The binding of this antibody to recombinant CCPG1 protein displayed a KD value of 3.9 x 10-7 whereas its binding to non-specific GST protein displayed a KD of 4.2 x 10-3.

Target description: Cell cycle progression protein 1 (UniProt: Q9ULG6; also known as Cell cycle progression restoration protein 8, CCPG1) is encoded by the CCPG1 (also known as CCP8, CPR8, KIAA1254) gene (Gene ID: 9236) in human. CCPG1 is a single-pass type II membrane protein that acts as an assembly platform for Rho protein signaling complexes. It is reported to mediate recruitment of endogenous Src kinase into Dbs-containing complexes and interacts with the Rho family member Cdc42. It limits guanine nucleotide exchange activity of MCF2L toward RHOA, which results in an inhibition of both its transcriptional activation ability and its transforming activity. However, it does not inhibit activity of MCF2L toward CDC42, or activity of MCF2 toward either RHOA or CDC42. CCPG1 also serves as an ER stress-inducible endoplasmic reticulum (ER)-phagy cargo receptor in mammals. It is shown to bind directly to ATG8 and FIP200 via distinct peptide motifs. It is reported to offer protection against ER luminal protein aggregation and consequent unfolded protein response hyperactivation and tissue injury of the exocrine pancreas. CCPG1 contains a cytoplasmic domain (aa 1-217), a transmembrane domain (aa 218-238), and a luminal domain (aa 239-757). This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
