Western Blotting Enhanced Validation - Recombinant Antibody Technology SH-SY5Y cell lysate was probed with Cat. No. ZRB1579, Anti-ISL-1, clone 1B11 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates ISL-1 (~42 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or ISL-1 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1579, Anti-ISL-1, clone 1B11 ZooMAb® Rabbit Monoclonal. The binding of this antibody to ISL-1 peptide displayed a KD of 5.6 x 10-8, whereas binding to non-specific control peptide displayed a KD of 1.6 x 10-4.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Specificity: Clone 1B11 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Insulin gene enhancer protein ISL-1. It targets an epitope within 21 amino acids within the LIM binding domain in the C-terminal region.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Insulin gene enhancer protein ISL-1 (UniProt: P61371; also known as Islet-1, ISL-1) is encoded by the ISL1 gene (Gene ID: 3670) in human. ISL-1 is a member of the LIM homeodomain family of transcription factors that contain two tandemly repeated cysteine/histidine-rich LIM domains that are involved in protein-protein interactions. It is a DNA-binding transcriptional activator that recognizes and binds to the consensus octamer binding site 5′-ATAATTAA-3′ in promoter of target genes. Its expression is observed in in subsets of neurons of the adrenal medulla and dorsal root ganglion, inner nuclear and ganglion cell layers in the retina, the pineal and some regions of the brain. It is shown to play a fundamental role in the gene regulatory network essential for retinal ganglion cell (RGC) differentiation. It is also involved in the specification of motor neurons in cooperation with LHX3 and LDB1. ISL-1 is also established as an important stem cell marker for cardiac progenitor cells in the developing heart. It is reported to bind to compacted chromatin and works in concert with the Brg1-Baf60c-based SWI/SNF complex to promote cardiac lineage-specific alterations in the chromatin landscape of genes that function in cardiac progenitors and cardiomyocyte structural genes. It is also expressed in a subpopulation of striatal progenitor cells that give rise to striatal neurons that belong to the striatonigral pathway.
