Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from HeLa cells (Lane 1), HEK293 cells (Lane 2), and NIH3T3 cells (Lane 3) were probed with Cat. Target description: Transducin-like enhancer protein 3 (UniProt: Q04726; also known as Enhancer of split Groucho-like protein 3, ESG3, TLE3) is encoded by the TLE3 (also known as KIAA1547) gene (Gene ID: 7090) in human. TLE3, a homotetrameric member of the Groucho/TLE family, is a transcriptional corepressor that binds to a number of transcription factors. Groucho/TLE proteins consist of a five-domain structures: a highly conserved Q domain that facilitates oligomerization of Gro/TLE molecules, a glycine/proline-rich (GP) domain that is essential for interaction of these proteins with histone deacetylases, a CcN domain that contains a nuclear localization sequence and CDC22 and casein kinase II phosphorylation sites, a serine/proline-rich domain, and a highly conserved WD40 domain that contains multiple tryptophan and aspartic acid tandem repeats. TLE3 is expressed in white adipose tissue and is shown to enhance the transcriptional activity of peroxisome proliferator-activated receptor (PPAR) and induce adipogenesis in preadipocytes. It also serves as a regulator of MyoD function during myogenesis. It interferes with MyoD function by disrupting the association between the basic helix-loop-helix domain of MyoD and E proteins. TLE3 is also shown to inhibit mitochondrial gene expression in beige adipocytes and conditional deletion of TLE3 in adipocytes is reported to promote mitochondrial oxidative metabolism and increase energy expenditure. It interacts with EBF2 and blocks its ability to promote the thermogenic transcriptional program. TLE3 is ubiquitinated by XIAP/BIRC4, but this ubiquitination does not affect its stability, nuclear localization, or capacity to tetramerize but inhibits its interaction with TCF7L2/TCF4. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
