Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from HeLa (Lane 1), PC12 (Lane 2), and NIH3T3 (Lane 3) cells were probed with Cat. No. ZRB1492, Anti-RAD51, clone 1M8 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates RAD51 (~37 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL human recombinant RAD51 protein (Run 1) or non-specific GST protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1492, Anti-RAD51, clone 1M8 ZooMAb® Rabbit Monoclonal. The binding of this antibody to recombinant RAD51 protein displayed a KD value of 1.0 x 10^-12 whereas its binding to non-specific control GST protein displayed a KD of 1.7 x 103.

Application: Anti-RAD51, clone 1M8 ZooMAb, Cat. No. ZRB1492, is a recombinant Rabbit monoclonal antibody that specifically targets RAD51 and is tested in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.
Application: Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RAD51 in HeLa and NIH3T3 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RAD51 in human testis tissue sections.

Affinity Binding Assay Analysis: 100 μg/mL from a representative lot detected RAD51 in RAD51 and GST protein showed a KD of 1.0 x 10^-12 whereas binding of ZRB1492 to non-specific GST protein showed a KD of 1.7 x 10^-3.

Target description: Nuclear factor erythroid 2-related factor 2 (UniProt: Q16236; also known as NF-E2-related factor 2, NFE2-related factor 2, Nrf-2, HEBP1, Nuclear factor, erythroid derived 2, like 2) is encoded by the NFE2L2 (also known as NRF2) gene (Gene ID: 4780) in human. Nrf2 is a cap n collar basic-region leucine zipper transcription factor that that heterodimerizes with small Maf proteins. It is involved in adaptation of cells to oxidative stress and electrophiles. It binds to antioxidant response (ARE) elements in the promoter regions of target genes and induces a set of cytoprotective genes that are involved in the synthesis of reduced glutathione (GSH), repair of oxidized protein thiols, scavenging of reactive oxygen species, and drug metabolism. It is cytosolic under unstressed conditions, but translocates into the nucleus upon induction by electrophilic agents. Under normal unstressed conditions it is rapidly turned over in a Keap-1 dependent manner through Cul3-Rbx1 ubiquitination and proteasomal degradation. Phosphorylation of Ser40 by protein kinase C in response to oxidative stress dissociates Nrf2 from its Keap-1 and promotes its translocation into the nucleus.
