Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from Jurkat cells (Lane 1), NIH3T3 cells (Lane 2), and L6 cells (Lane 3) were probed with Cat. No. ZRB1458, Anti-RAD21, clone 2H12 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates RAD21 (~105 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 mg/mL of recombinant RAD21 protein fragment (Run 1) or non-specific GST protein (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1458, Anti-RAD21, clone 2H12 ZooMAb® Rabbit Monoclonal. The binding of this antibody to RAD21 fragment displayed a KD value of 9.0 x 108 whereas its binding to non-specific GST displayed a KD of 2.0 x 105.

Target description: Double-strand-break repair protein rad21 homolog (UniProt: O60216; also known as hHR21, Nuclear matrix protein 1, NXP-1, SCC1 homolog) is encoded by the RAD21 (also known as HR21, KIAA0078, NXP1, SCC1) gene (Gene ID: 5885) in human. RAD21 is a double-strand-break repair protein rad21 homolog. It is a component of the cohesion complex and is integral to chromosome segregation, error-free DNA repair, and prevention of inappropriate recombination between repetitive regions. It is cleaved by separase/ESPL1 at the onset of anaphase and this cleavage is required for sister chromatid separation and cytokinesis. The cohesin complex also plays a role in spindle pole assembly during mitosis. RAD21 expression in colorectal cancer is associated with aggressive disease especially in KRAS mutant tumors and resistance to chemoradiotherapy. Mutations in RAD21 gene have been linked to Cornelia de Lange syndrome that is characterized by facial dysmorphisms, abnormal hands and feet, growth delay and cognitive retardation. Some mutations can also cause Mungan syndrome that leads to neuromyopathy, chronic intestinal pseudo-obstruction and a variety of cardiac valve defects.
