Western Blotting Enhanced Validation-Recombinant Antibody Technology HepG2 (Lane 1), A549 (Lane 2) cell lysates were probed with Cat. No. ZRB1456, Anti-Aquaporin-9, clone 1E4 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Aquaporin-9 (~26 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of Aquaporin-9 peptide (Run 1) or a non-specific control peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1456, Anti-Aquaporin-9, clone 1E4 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Aquaporin-9 peptide displayed a KD value of 8.0 x 106 whereas its binding to non-specific peptide displayed a KD of 1.5 x 103.

Target description: Aquaporin-9 (UniProt: O43315; also known AQP-9, Aquaglyceroporin-9, Small solute channel 1) is encoded by the AQP9 (also known as SSC1) gene (Gene ID: 366) in human. Aquaporins are a family of small transmembrane proteins that facilitate osmotically driven water transport. Aquaporin-9 is a multi-pass membrane protein that forms a water channel with a broad specificity. It also mediates passage of a wide variety of small, non-charged solutes including purines, and pyrimidines. It is highly expressed in peripheral leukocytes and its expression is also reported in liver, lung, and spleen. Aquaporins contain two tandem repeats each containing three membrane-spanning domains and a pore-forming loop with the signature motif Asn-Pro-Ala (NPA; aa 84-86 and 216-218). This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
