Immunostaining was performed using a 1:1,000 dilution of Cat. No. ZRB1444, Anti-PEA15, clone 2J13 ZooMAb® Rabbit Monoclonal. Reactivity was detected using an Goat Anti-Rabbit IgG and HRP-DAB. Cytoplasmic/membranous staining was observed in subsets of neurons, glial and endothelial cells, as well as neuropil of human cerebral cortex.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from NIH3T3 (Lane 1), C6 (Lane 2), and SH-SY5Y (Lane 3) were probed with Cat. No. ZRB1444, Anti-PEA15, clone 2J13 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates PEA15 (~15 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of non-specific GST protein (Run 2) or recombinant PEA15 protein (Run 1) were analyzed by affinity binding assay using Cat. No. ZRB1444, Anti-PEA15, clone 2J13 ZooMAb® Rabbit Monoclonal. The binding of this antibody to recombinant PEA15 protein displayed a KD value of 2.7 x 105 whereas its binding to non-specific protein displayed a KD of 2.4 x 103.

Target description: Neurofilament light polypeptide (UniProt: P07196; also known as NF-L, 68 kDa neurofilament protein, Neurofilament triplet L protein) is encoded by the NEFL (also known as NF68, NFL) gene (Gene ID: 4747) in human. Neurofilaments (~ 10 nm in diameter) are intermediate filaments that serve as major elements of the cytoskeleton supporting the axon cytoplasm. They are the most abundant fibrillar components of the axon, being on average 3-10 times more frequent than axonal microtubules. They are built from three intertwined protofibrils, which are themselves composed of two tetrameric protofilament complexes of monomeric proteins. The neurofilament triplet proteins (68/70, 160, and 200 kDa) occur in both the central and peripheral nervous system and are usually neuron specific. The 68/70 kDa NF-L protein can self-assemble into a filamentous structure, however the 160 kDa NF-M and 200 kDa NF-H proteins require the presence of the 68/70 kDa NF-L protein to co-assemble. NF-L is the most abundant of the three neurofilament proteins. Mutations in NEFL gene are known to cause Charcot-Marie-Tooth disease 1F, a disorder of the peripheral nervous system characterized by progressive weakness and atrophy.
