Western Blotting Enhanced Validation-Recombinant Antibody Technology Human (Lane 1), Rat (Lane 2), and Mouse Skin (Lane 3) tissue lysates were probed with Cat. No. ZRB1441, Anti-Cytokeratin-14, clone 1D15 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Cytokeratin-14 (~52 kDa).

Target description: Keratin, type I cytoskeletal 14 (UniProt: P02533; also known as Cytokeratin-14, CK-14, Keratin-14, K14) is encoded by the KRT14 gene (Gene ID: 3861) in human. The keratins are the intermediate filament proteins of epithelia that display high degree of molecular diversity. They are heteropolymeric filaments formed by pairing of type I and type II keratins that are expressed in a highly specific patterns depending on the epithelial type and stage of cellular differentiation. Keratins contain a central rod domain of about 310 amino acids with alpha-helical conformation bordered by non-helical head and tail domains of variable length. The head domain consists of subdomains V1 and H1. The central alpha helical rod domain is composed of subdomains 1A, 1B, 2A, and 2B connected by the linkers L1, L12, and L2. The tail domain then consists of subdomains H2 and V2. They provide stability between epithelial cells and their attachment to basement membrane. Keratins also serve as tumor markers and epithelial differentiation markers. Cytokeratin-14 is detected in the basal layer, lowered within the more apically located layers specifically in the stratum spinosum, stratum granulosum, but is not detected in stratum corneum. It is strongly expressed in the outer root sheath of anagen follicles. It is present in nucleus and cytoplasm and is expressed in a filamentous pattern. Target description: Proliferating cell nuclear antigen (UniProt: P12004; also known as PCNA) is encoded by the PCNA gene (Gene ID: 5111) in human. PCNA is a homotrimeric nuclear protein that serves as an auxiliary protein of DNA polymerase delta and is involved in the control of eukaryotic DNA replication by increasing the polymerase′s processability during elongation of the leading strand. It Induces a robust stimulatory effect on the 3′-5′ exonuclease and 3′-phosphodiesterase, but not apurinic-apyrimidinic (AP) endonuclease, APEX2 activities. It is reported to colocalize with CREBBP, EP300 and POLD1 to sites of DNA damage. It acts as a loading platform to recruit DDR proteins that allow completion of DNA replication after DNA damage and promote post-replication repair. PCNA can undergo phosphorylation on tyrosine 211 by EGFR that stabilizes chromatin-associated PCNA. Following DNA damage, it can undergo monoubiquitination to stimulate direct bypass of DNA lesions by specialized DNA polymerases or polyubiquitination to promote recombination-dependent DNA synthesis across DNA lesions by template switching mechanisms. Mutation in PCN gene are known to cause Ataxia-telangiectasia-like disorder 2 (ATLD2) that is characterized by developmental delay, ataxia, hearing loss, and short stature.
