Western Blotting Enhanced Validation-Recombinant Antibody Technology Human Lymph Node (Lane 1) and Human Lung (Lane 2) tissue lysates were probed with Cat. No. ZRB1440, Anti-MRC1, clone 1C9 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates MRC1 (~180 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 mg/mL of non-specific peptide (Run 2) or MRC1 peptide (Run 1) were analyzed by affinity binding assay using Cat. No. ZRB1440, Anti-MRC1, clone 1C9 ZooMAb® Rabbit Monoclonal. The binding of this antibody to MRC1 peptide displayed a KD value of 4.6 x 108 whereas its binding to non-specific peptide displayed a KD of 1.5 x 105.
Flow Cytometry Enhanced Validation-Recombinant Antibody Technology Staining of one million RAW 264.7 cells was performed using 0.01 μg of Cat. No. ZRB1440, Anti-MRC1, clone 1C9 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-Rabbit secondary antibody.

Target description: Macrophage mannose receptor 1 (UniProt: P22897; also known as MMR, C-type lectin domain family 13 member D, C-type lectin domain family 13 member D-like, Human mannose receptor, hMR, Macrophage mannose receptor 1-like protein 1, CD206, MRC1) is encoded by the MRC1 (also known as CLEC13D, CLEC13DL, MRC1L1) gene (Gene ID: 4360) in human. MRC1 is a single-pass type I membrane protein that mediates the endocytosis of glycoproteins by macrophages. It also functions in phagocytosis and plays a role in immune homeostasis by scavenging unwanted mannoglycoproteins. It is expressed on the surface of macrophages, immature dendritic cells and liver sinusoidal endothelial cells. It is a C-type lectin with eight C-type lectin domains and can bind both sulfated and non-sulfated polysaccharide chains. MRC1 is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to generate the mature form that contains a large extracellular domain (aa 19-1389), a transmembrane domain (aa 1390-13410), and a cytoplasmic tail (aa 1411-1456). The extracellular region includes an N-terminal cysteine-rich domain that binds glycoproteins bearing sulfated sugars that terminate in 4-SO4GalNAc, a fibronectin II (FNII) domain, and eight carbohydrate recognition domains that bind sugars like mannose and fucose with high affinity. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
