Western Blotting Enhanced Validation-Recombinant Antibody Technology HeLa cell lysate was probed with Cat. No. ZRB1439, Anti-MICB, clone 2K9 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates MICB (~55 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 mg/mL of non-specific peptide (Run 1) or MICB peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1439, Anti-MICB, clone 2K9 ZooMAb® Rabbit Monoclonal. The binding of this antibody to MICB peptide displayed a KD value of 1.0 x 106 whereas its binding to non-specific peptide displayed a KD of 4.3 x 103.
Flow Cytometry Enhanced Validation-Recombinant Antibody Technology Staining of one million HeLa cells was performed using 0.1 μg of Cat. No. ZRB1439, Anti-MICB, clone 2K9 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-Rabbit secondary antibody.

Target description: MHC class I polypeptide-related sequence B (UniProt: Q29980; also known as MIC-B, MICB) is encoded by the MICB (also known as PERB11.2) gene (Gene ID: 4277) in human. MICB is a single-pass type I membrane protein that is widely expressed and acts as a stress-induced self-antigen. It is widely expressed with the exception of the central nervous system. It is also expressed in many, but not all, epithelial tumors of lung, breast, kidney, ovary, prostate and colon. It is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 23-309), a transmembrane domain (aa 310-330), and a cytoplasmic domain (aa 331-383). MICB can be induced by heat shock, oxidative stress, retinoic acid, interferon alpha, and 5-aza-2′-deoxycytidine. Its induction by interferon alpha is impaired in subjects with chronic hepatitis C virus infection. It is recognized by gamma-delta T cells and serves as ligands for the KLRK1/NKG2D receptor. Its binding to KLRK1 leads to cell lysis. Mutations in MICB gene have been linked to Rheumatoid arthritis that is characterized by inflammatory changes in the synovial membranes and articular structures, along with widespread fibrinoid degeneration of the collagen fibers in mesenchymal tissues.
