Western Blotting Enhanced Validation - Recombinant Antibody Technology Mouse Brain (Lane 1), Mouse Brain nuclear extract (Lane 2), and Rat Brain nuclear extract (Lane 3) tissue lysates were probed with Cat. No. ZRB1436, Anti-Olig2, clone 1K2 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Olig2 (~41 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology Affinity binding assay: 100 mg/mL of non-specific peptide (GST; Run 1) or recombinant Olig2 (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1436, Anti-Olig2, clone 1K2 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Olig2 displayed a KD value of 2.7 x 10-7 whereas its binding to GST displayed a KD of 5.1 x 10-2.

Target description: Oligodendrocyte transcription factor 2 (UniProt: Q13516; also known as Oligo2, Class B basic helix-loop-helix protein 1, bHLHb1, Class E basic helix-loop-helix protein 19, bHLHe19, Protein kinase C-binding protein 2, Protein kinase C-binding protein RACK17, Olig2) is encoded by the OLIG2 (also known as BHLHB1, BHLHE19, PRKCBP2, RACK17) gene (Gene ID: 10215) in human. Olig2 is a is a basic helix-loop-helix (bHLH) transcription factor that is required for oligodendrocyte and motor neuron specification in the spinal cord, as well as for the development of somatic motor neurons in the hindbrain. It is expressed in oligodendrocytes and stronger expression is reported in oligodendrogliomas. It functions together with ZNF488 to promote oligodendrocyte differentiation and cooperates with OLIG1 to establish the pMN domain of the embryonic neural tube. A chromosomal aberration involving OLIG2 is reported to cause a form of T-cell acute lymphoblastic leukemia (T-ALL). OLIG2 expression is induced by sonic hedgehog and fibroblast growth factor during development. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
