Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from HEK293 (Lane 1), C6 (Lane 2), and C2C12 (Lane 3) cells were probed with Cat. No. ZRB1432, Anti-p-Myosin LC 2 (Ser19), clone 2C10 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates p-Myosin LC 2 (Ser19) (~19 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-phosphorylated myosin light chain 2 control peptide (Run 1) or phosphorylated myosin light chain 2 (Ser19) peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1432, Anti-p-Myosin LC 2 (Ser19), clone 2C10 ZooMAb® Rabbit Monoclonal. The binding of this antibody to phosphorylated myosin light chain 2 (Ser19) peptide displayed a KD of 6.0 x 10-6, whereas binding to non-phosphorylated myosin control peptide displayed a KD of 3.6 x 10-3.

Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. Target description: Myosin regulatory light polypeptide 9 (UniProt: P24844; also known as 20 kDa myosin light chain, LC20, MLC-2C, Myosin RLC, Myosin regulatory light chain 2 smooth muscle isoform, Myosin regulatory light chain 9, Myosin regulatory light chain MRLC1) is encoded by the MLY9 (also known as MLC2, MRLC1, MYRL2) gene (Gene ID: 10398) in human. Myosins are highly conserved, ubiquitously expressed proteins that form regular bipolar thick filaments that together with actin thin filaments constitute the fundamental contractile unit of skeletal and cardiac muscle. Along with actin they serve as motor molecules with ATPase activity that is essential for muscle contraction. Muscle myosin is a hexameric protein that consists of 2 heavy chain subunits (MHC), 2 light chain subunits (MLC) and 2 regulatory light chain subunits (MLC-2). MLC-2C plays an important role in regulation of both smooth muscle and non-muscle cell contractile activity via its phosphorylation. It is phosphorylated at threonine 18 and serine 19 by myosin light chain kinase (MLCK) in a Ca2+/CaM-dependent manner. Rho kinase (ROCK) is also known to phosphorylates MLC-2C at serine 19. Phosphorylation increases the actin-activated myosin ATPase activity and thereby regulates the contractile activity. It is required to generate the driving force in the migration of the cells. Phosphorylation is also required for myotube formation.
