Target description: Nucleolin (UniProt: P19338; also known as Protein C23) is encoded by the NCL gene (Gene ID: 4691) in human. Nucleolin is a multifunctional phosphoprotein that is ubiquitously distributed in the nucleolus, nucleus and cytoplasm of the cell. It is one of the most abundant non-ribosomal proteins that accounts for about 10% of the protein content within the nucleolus. It is found associated with intra-nucleolar chromatin and pre-ribosomal particles. It plays important roles in various steps of ribosomal synthesis, including the transcription of rDNA repeats, the modification and processing of pre-rRNA, the assembly of pre-ribosomal particles and nuclear-cytoplasmic transport of ribosomal proteins and subunits. Nucleolin contains three structural and multifunctional domains: the N-terminal domain contains several acidic stretches; two to four RNA-binding domains (RNA recognition motifs or RRM) in the central region; and a glycine/arginine-rich domain (GAR) domain at the C-terminus. Nucleolin is shown to be highly phosphorylated. It undergoes extensive phosphorylation by casein kinase 2 at interphase and by CDC2 during mitosis. It has been reported that Nucleolin phosphorylation at interphase is correlated with active rRNA transcription. Although the mammalian Nucleolin has a predicted molecular weight of about 77 kDa, its apparent molecular weight lies between 100 and 110 kDa due to highly phosphorylated amino acids in the N-terminal domain.
