Western Blotting Enhanced Validation - Recombinant Antibody Technology Human lymph node (Lane 1) and mouse spleen (Lane 2) tissue lysates were probed with Cat. No. ZRB1412, Anti-CD25 (IL2-RA), clone 1K6 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates CD25 (IL2-RA) (~35 kDa).
Flow Cytometry Enhanced Validation - Recombinant Antibody Technology Staining of one million human PBMC was performed using 1 μg of Cat. No. ZRB1412, Anti-CD25 (IL2-RA), clone 1K6 ZooMAb® Rabbit Monoclonal and visualized with a Donkey Anti-Rabbit secondary antibody and 1 μg of Rabbit IgG isotype control. Histogram represents the gated R1 population shown in the dot plot.

Application: Anti-CD25 (IL2-RA), clone 1K6 ZooMAb, Cat. No. ZRB1412, is a recombinant Rabbit monoclonal antibody that targets CD25 (IL2-RA) and is tested for use in Flow Cytometry, Immunohistochemistry (Paraffin), and Western Blotting.
Application: Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD25 (IL2-RA) in human lymph node tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD25 (IL2-RA) in human tonsil tissue sections.

Flow Cytometry Analysis: 1μg from a representative lot detected CD25 (IL2-RA) in one million human peripheral blood mononuclear cells (PBMC).

Target description: Interleukin-2 receptor subunit alpha (UniProt: P01589; also known as IL-2 receptor subunit alpha, IL-2-RA, IL-2R subunit alpha, IL2-RA, TAC antigen, p55, CD25) is encoded by the IL2RA gene (Gene ID: 3559) in human. IL-2-RA is a non-covalent dimer of an alpha (CD25), beta unit (CD122). IL-2-RA is involved in the regulation of immune tolerance and controls the activity of T cells. IL-2RA can exist in three different forms; a high affinity dimer, an intermediate affinity monomer of beta subunit, and a low affinity monomer of alpha subunit. The high and intermediate affinity forms can also associate with a gamma subunit (CD132). CD25 is rapidly up-regulated upon activation of T cells. Activated T cells, regulatory T cells (Tregs) and NK cells express high levels of CD25, and high affinity expression of IL-2-RA is largely limited to these cells. CD25 is a single-pass type I membrane protein that is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 22-240), a transmembrane domain (aa 241-250), and a short cytoplasmic tail (aa 260-272). It also contains two Sushi domains (aa 22-84 and 123-186). Mutations in IL2RA gene have been linked to insulin-dependent diabetes mellitus and immunodeficiency with lymphoproliferation that leads to recurrent viral, fungal, and bacterial infections. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
