Target description: Unconventional myosin-Ic (UniProt: O00159; also known as Myosin I beta, MMI-beta, MMIb, MYO1C) is encoded by the MYO1C gene (Gene ID: 4641) in human. Myosins are actin-based motor molecules with ATPase activity essential for muscle contraction. Myosins form regular bipolar thick filaments that together with actin thin filaments constitute the fundamental contractile unit of skeletal and cardiac muscle. Unconventional myosins serve in intracellular movements. Their highly divergent tails are presumed to bind to membranous compartments. MYO1C, a single-headed class I myosin, localized to the cell surface. It is associated with cholesterol-enriched lipid rafts and enhances their recycling from intracellular compartments to the cell surface. It is involved in glucose transporter recycling in response to insulin by regulating movement of intracellular GLUT4-containing vesicles to the plasma membrane. It is reported to colocalize with RNA polymerase II in the nucleus and with RNA polymerase I in nucleoli. In the nucleolus, it is localized predominantly in the dense fibrillar component (DFC) and in granular component. The MYO1C gene produces three alternatively spliced isoforms that differ only in their N-terminal regions. Although they share identical motor domains, three calmodulin-binding IQ motifs containing a nuclear localization signal, and a membrane-binding tail domain, each isoform has different function and nuclear cytoplasm partitioning. These isoforms display both specific and overlapping nuclear and cytoplasmic functions and have different expression levels. Isoform 3 of MYO1C is involved in regulation of transcription and associates with transcriptional active ribosomal genes and cooperates with the WICH chromatin-remodeling complex to facilitate transcription. It is shown to be essential for the formation of the first phosphodiester bond during transcription initiation. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
