Immunocytochemistry Enhanced Validation - Recombinant Antibody Technology Immunofluorescent analysis of U-2 OS cell line was performed using a 1:100 dilution of Cat. No. ZRB1371, Anti-NCOA1, clone 5B9 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488 (Green). Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.

Western Blotting Enhanced Validation - Recombinant Antibody Technology HEK293 cell lysate was probed with Cat. No. ZRB1371, Anti-NCOA1, clone 5B9 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates NCOA1 (~175 kDa).
Affintiy Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of human recombinant NCOA1 (Run 1) and a non-specific protein (GST; Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1371, Anti-NCOA1, clone 5B9 ZooMAb® Rabbit Monoclonal. The binding of this antibody to recombinant NCOA1 displayed a KD value of 6.9 x 10-7 whereas its binding to non-specific peptide displayed a KD of 3.9 x 10-3.

Target description: Nuclear receptor coactivator 1 (UniProt: Q15788; also known as EC:2.3.1.48, NCoA-1, Class E basic helix-loop-helix protein 74, bHLHe74, Protein Hin-2, RIP160, Renal carcinoma antigen NY-REN-52, Steroid receptor coactivator 1, SRC-1) is encoded by the NCOA1 (also known as BHLHE74, SRC1) gene (Gene ID: 8648) in human. NCOA1 I a member of the SRC/p160 nuclear receptor coactivator family that is widely expressed. It serves as a nuclear receptor coactivator that directly binds nuclear receptors and plays crucial roles in the regulation of gene expression mediated by a wide spectrum of steroid receptors such as androgen receptor, estrogen receptor alpha, and beta. It also displays histone acetyltransferase activity toward H3 and H4. NCOA1 contains seven LXXLL motifs of which motifs 3, 4 and 5 are essential for the association with nuclear receptors and motif 7 increases the affinity for steroid receptors in vitro. It is recruited to AF2 domains of ligand-activated nuclear receptors (NR) through LXXLL motifs located in a central NR interaction domain. It contains both a basic-helix-loop-helix (bHLH) domain (aa 23-80) and a periodicity/aryl hydrocarbon receptor nuclear translocator (ARNT)/single-minded (PAS) domain (aa 109-180). The bHLH-PAS domain houses a nuclear localization sequence and interacts with the Baf57 subunit of the SWI/SNF ATP-dependent chromatin remodeling complex. It also has a serine/threonine-rich region within the -terminal half (between amino acids 300 and 600) that allows for various posttranslational modifications. Dysregulation of NCOA1 has been linked to a variety of cancers and its elevated levels are reported to enhance breast cancer cell proliferation and metastasis. A chromosomal aberration involving NCOA1 has also been shown to be a cause for rhabdomyosarcoma. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
