No. ZRB1344, Anti-Tec, clone 19M23 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488 (Green). Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the plasma membrane.

Western Blotting Enhanced Validation - Recombinant Antibody Technology K562 cell lysate was probed with Cat. No. ZRB1344, Anti-Tec, clone 19M23 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Tec (~65 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of Tec peptide (Run 1) or non-specific peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB11344, Anti-Tec, clone 19M23 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Tec peptide displayed a KD value of 1.2 x 10-5 whereas its binding to non-specific peptide displayed a KD of 6.3 x 10-3.
ELISA Enhanced Validation - Recombinant Antibody Technology Serial dilutions of 0.5 μg/mL of peptide immunogens (Yellow) was analyzed by sandwich ELISA using Cat. No. ZRB1344, Anti-Tec, clone 19M23 ZooMAb® Rabbit Monoclonal using a Goat Anti-Rabbit secondary antibody. This antibody specifically detected Tec, but not any peptides from other proteins.

Target description: Tyrosine-protein kinase Tec (UniProt: P42680; also known as EC:2.7.10.2, Tec) is encoded by the TEC (also known as PSCTK4) gene (Gene ID: 7006) in human. The Tec family of protein kinases consists of five members, Tec, Btk, Itk/Emt/Tsk, Bmx and Txk/Rlk. They are abundantly expressed in hematopoietic cells and participate in the growth and differentiation of blood cells. Tec is a peripheral membrane protein that serves as a non-receptor tyrosine kinase that contributes to signaling from many receptors and participates as a signal transducer in multiple downstream pathways, including regulation of the actin cytoskeleton. It is activated by tyrosine phosphorylation by a wide range of cytokine stimulations. It regulates the development, function and differentiation of conventional T-cells and nonconventional NKT-cells and is required for TCR-dependent IL2 gene induction. Following B-cell or T-cell receptors activation by antigen, it is shown to translocate to the plasma membrane through its PH domain (aa 4-111). Tec is reported to serve as a central regulator of the noncanonical caspase-8 inflammasome that is involve in the assembly and activation of the caspase-8 inflammasome in response to fungal pathogens but is not involved in anti-bacterial immune responses.
