Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Specificity: Clone 2G22 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Methyltransferase-like protein 14 (METTL14). It targets an epitope within 16 amino acids from the C-terminal half.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: N6-adenosine-methyltransferase non-catalytic subunit (UniProt: Q9HCE5; also known as Methyltransferase-like protein 14, hMETTL14) is encoded by the METTL14 (also known as KIAA1627) gene (Gene ID: 57721) in human. METTL14 forms a heterodimer with METTL3 to form a N6-methyltransferase complex that methylates adenosine residues at the N6 position of some mRNAs and regulates circadian clock, differentiation of embryonic stem cells, and cortical neurogenesis. This METTL14-METTL3 is an antiparallel heterodimer with active methyltransferase activity. In this heterodimer METTL14 constitutes the RNA-binding scaffold that recognizes the substrate rather than the catalytic core. N6-methyladenosine (m6A) is one of the most abundant post-transcriptional, reversible modification of RNA in eukaryotic cells. It is a developmentally regulated RNA modification that is dynamically modified. It is an important physiological regulator involved in cell renewal and differentiation and may function in post-transcriptional fine-tuning of gene expression and may provide a faster means of maximizing gene expression.
