Target description: Tubulin alpha-4A chain (UniProt: P68366; also known as Alpha-tubulin 1, Testis-specific alpha-tubulin, Tubulin H2-alpha, Tubulin alpha-1 chain) is encoded by the TUBA4A (also known as TUBA1) gene (Gene ID: 7277) in human. Microtubules are cytoskeletal filaments composed of alpha-/beta-tubulin heterodimers that self-assemble head-to-tail to form protofilaments and laterally to form a hollow tube. Alpha-beta heterodimers associate head-to-tail to form protofilaments running lengthwise along the microtubule wall with the beta-tubulin subunit facing the microtubule plus end conferring a structural polarity. Tubulin binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non-exchangeable site on the alpha chain. Various post-translational modifications, including acetylation, polyglutamylation, polyglycylation, detyrosination, phosphorylation, and palmitoylation, can regulate the polymerization properties of tubulins and/or their interactions with microtubule associated proteins and motor proteins. The GNAT lysine acetyltransferase family member MEC-17 (or alpha-tubulin acetyltransferase/alpha TAT) is reported to catalyze lysine 40 acetylation, while HDAC6 and sirtuin2 (SIRT2) are known to catalyze tubulin deacetylation. Acetylation of alpha chains at lysine 40 is located inside the microtubule lumen. This modification has been correlated with increased microtubule stability, intracellular transport and ciliary assembly.
