Target description: G1/S-specific cyclin-D1 (UniProt: P24385; also known as B-cell lymphoma 1 protein, BCL-1, BCL-1 oncogene, PRAD1 oncogene) is encoded by the CCND1 (also known as BCL1, PRAD1) gene (Gene ID: 595) in human. Cyclin D1 is an considered to be an essential regulator of the G1-S transition in response to growth factor stimulation in somatic cells. Together with cyclin dependent kinases 4 and 6 (CDK4 and CDK6), it forms active complexes that are shown to promote cell cycle progression by phosphorylating and inactivating the retinoblastoma protein (RB). Phosphorylation of RB1 allows dissociation of E2F from the RB/E2F complex and the subsequent transcription of E2F target genes that are responsible for the progression through the G1 phase. Cyclin D1 levels are reported to rise early in G1 and continue to accumulate until the G1/S-phase boundary when its levels rapidly decline. It exhibits a short half-life of about 24 minutes and is readily degraded in the 26S proteasome. Cyclin D1 is shown to repress DNA replication by binding to proliferating cell nuclear antigen (PCNA) and Cdk2. The binding of cyclin D1 to PCNA directly inhibits DNA synthesis. The overexpression of cyclin D1 and its deregulated degradation have been linked to the development and progression of various cancers. A chromosomal aberration involving CCND1 gene is reported to cause B-lymphocytic malignancy.
