Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from Human umbilical vein endothelial cells (HUVEC; Lane 1) and Human Lung tissue (Lane 2) were probed with Cat. No. ZRB1297, Anti-Endoglin, clone 1I15 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Endoglin (~90 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or Endoglin peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1297, Anti-Endoglin, clone 1I15 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Endoglin peptide displayed a KD of 3.8 x 10-7 whereas binding to non-specific control peptide displayed a KD of 3.8 x 10-4.
Flow Cytometry Enhanced Validation - Recombinant Antibody Technology Staining of one million Human umbilical vein endothelial cells (HUVEC) was performed using 1 μg of Cat. No. ZRB1297, Anti-Endoglin, clone 1I15 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-Rabbit IgG secondary antibody.

| Target description: | Endoglin (UniProt: P17813; also known as CD105) is encoded by the ENG (also known as END) gene (Gene ID: 2022) in human. Endoglin is a single-pass type I, disulfide-linked, membrane glycoprotein that is detected on placenta and umbilical vein endothelial cells. It can also form heterodimeric complex with the signaling receptors for TGFb. It is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 26-586), a transmembrane domain (aa 587-611), and a cytoplasmic domain (aa 612-658). Endoglin also contains a zona pellucida (ZP) domain (aa 363-533) that mediates its dimerization. Endoglin is reported to play an important role in the regulation of angiogenesis. It is also required for normal structure and integrity of adult vasculature. It regulates the migration of vascular endothelial cells and regulates the change in shape of endothelial cells in response to blood flow thereby causing vascular remodeling and establishment of normal vascular morphology during angiogenesis. It has been also reported to play a role in the binding of endothelial cells to integrins and/or other RGD receptors. Endoglin also serves as a co-receptor for TGFb and is involved in the TGFb/BMP signaling cascade leading to the activation of SMAD transcription factors. Mutations in ENG gene have been linked to hereditary hemorrhagic telangiectasia that is characterized by recurrent epistaxis and gastro-intestinal hemorrhage.
