Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the endosomes.

Western Blotting Enhanced Validation - Recombinant Antibody Technology Mouse brain membrane preparation was probed with Cat. No. ZRB1283, Anti-ASTN2, clone 2E8 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates ASTN2 (~105 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or ASTN2 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1283, Anti-ASTN2, clone 2E8 ZooMAb® Rabbit Monoclonal. The binding of this antibody to ASTN2 peptide displayed a KD of 1.0 x 10-12 whereas binding to non-specific control peptide displayed a KD of 2.3 x 10-3.
Flow Cytometry Enhanced Validation - Recombinant Antibody Technology Staining of one million A431 cells was performed using 1 μg of Cat. No. ZRB1283, Anti-ASTN2, clone 2E8 ZooMAb® Rabbit Monoclonal (Yellow histogram) or the equivalent amount of a Rabbit IgG isotype control (Grey histogram), followed by a PE-conjugated Donkey Anti-Rabbit IgG secondary antibody.

Tested Applications

Affinity Binding Assay:: A representative lot of this antibody bound ASTN2 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Flow Cytometry Analysis: 1 μg from a representative lot detected ASTN2 in one million A431 cells.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ASTN2 in A431 cells.

Target description: Astrotactin-2 (UniProt: O75129; also known as ASTN2) is encoded by the ASTN2 (also known as KIAA0634) gene (Gene ID: 23245) in human. Astrotactin-2 is a multi-pass membrane protein that is synthesized with a signal peptide (aa 1-49), which is subsequently cleaved off and the mature form has two lumenal domains (aa 50-206 and 456-1339), two transmembrane domains (aa 207-227 and 435-455) and one cytoplasmic domain (aa 228-434). It also contains three EGF-like domains and one fibronectin type III domain. Its highest expression is observed in the cerebellum and lower levels of expression are shown in hippocampus, cortex, and olfactory bulb. It localizes mostly to vesicles inside neural cells that may give a punctate antibody staining pattern in the soma and along neuronal processes. Astrotactin-2 interacts with Astrotactin-1 and mediates its recycling to the anterior pole of the cell membrane in migrating neurons. It is shown to be a key regulator of dynamic trafficking of synaptic proteins. It associates with recycling and degradative vesicles in cerebellar neurons and promotes endocytic trafficking and degradation of synaptic proteins. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
