Target description: | Myelin-associated glycoprotein (UniProt: P20916; also known as Siglec-4a, MAG) is encoded by the MAG (also known as GMA) gene (Gene ID: 4099) in human. MAG is a single-pass type I membrane protein that is expressed in myelin-forming cells when oligodendrites initiate contact with axons. It is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 20-516), a transmembrane domain (aa 517-536), and a cytoplasmic domain (aa 518-626). MAG is a member of the immunoglobulin superfamily that is found in both peripheral and CNS myelin, where it plays a role in the formation and maintenance of myelin sheath. It is not required for initial myelination, but seems to play a role in the maintenance of normal axon myelination. MAG is reported to protect motoneurons against apoptosis and after injury. Protection against apoptosis is probably mediated via interaction with neuronal reticulon receptor 4R (RTN4R) and Reticulon-4 receptor-like 2 protein (RTN4RL2). It is also considered to be important in maintenance of myelin with aging.
