Immunocytochemistry Enhanced Validation - Recombinant Antibody Technology Immunofluorescent analysis of NIH 3T3 cells was performed using a 1:100 dilution of Cat. No. ZRB1243, Anti-FGFR1, clone 2J21 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488. Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the membrane receptor tyrosine kinase.

Western Blotting Enhanced Validation - Recombinant Antibody Technology NIH3T3 cell lysate was probed with Cat. No. ZRB1243, Anti-FGFR1, clone 2J21 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates FGFR1 (~80 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or FGFR1 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1243, Anti-FGFR1, clone 2J21 ZooMAb® Rabbit Monoclonal. The binding of this antibody to FGFR1 peptide displayed a KD of 6.9 x 10-6, whereas binding to non-specific control peptide displayed a KD of 3.2 x 10-3.

Target description: Fibroblast growth factor receptor 1 (UniProt: P11362; also known as EC:2.7.10.1, FGFR-1, Basic fibroblast growth factor receptor 1, BFGFR, bFGF-R-1, Fms-like tyrosine kinase 2, FLT-2, N-sam, Proto-oncogene c-Fgr, CD331, FGFR1) is encoded by the FGFR1 (also known as BFGFR, CEK, FGFBR, FLG, FLT2, HBGFR) gene (Gene ID: 2260) in human. FGFR1 is a monomeric single-pass type I membrane protein that undergoes homodimerization upon ligand binding. It is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 22-376), a transmembrane domain (aa 377-397), and a cytoplasmic domain (aa 398-822). It serves as a cell-surface receptor for fibroblast growth factors and plays an essential role in the regulation of embryonic development, cell proliferation, differentiation and migration. It is shown to be essential for normal mesoderm patterning and correct axial organization during embryonic development. FGFR1 is present in an inactive conformation in the absence of bound ligand and undergoes homodimerization and activation upon ligand binding and sequential autophosphorylation on tyrosine residues. After ligand binding, both receptor and ligand are rapidly internalized. Initial autophosphorylation at tyrosine 653 is shown to increase its tyrosine kinase activity by a factor of 50 to 100 and subsequently it is phosphorylated on tyrosine 583, 463, 766, 585, and 654, which causes a 10-fold increase in its kinase activity. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
