Target description: | Caspase-1 (UniProt: P29466; also known as EC:3.4.22.36, CASP-1, Interleukin-1 beta convertase, IL-1BC, Interleukin-1 beta-converting enzyme, ICE, IL-1 beta-converting enzyme, p45) is encoded by the CASP1 (also known as IL1BC, IL1BCE) gene (Gene ID: 834) inn human. Caspse-1 is a thiol protease that is synthesized as a single chain zymogen, which is further processed to create active caspase-1. The zymogen has two propeptides (aa 1-119 and 298-316), that are subsequently cleaved off to generate p20 (aa 120-297) and p10 (aa 317-404) subunits. The mature enzyme consists of two anti-parallel arranged heterodimers, each formed by a p20 and a p10 subunit. The p20 subunit can also form a heterodimer with the epsilon isoform which then has an inhibitory effect. It displays a strict requirement for an aspartic acid residue at position P1 and has a preferred cleavage sequence of Tyr-Val-Ala-Asp. Caspase-1 cleaves IL-1 beta between an aspartic acid and alanine, releasing the mature cytokine which is involved in a variety of inflammatory processes. It is an important component of the inflammasome that during DNA virus infection controls antiviral immunity through cleavage of cGMP-AMP synthase (cGAS), rendering it inactive. Caspase-1 is expressed in larger amounts in spleen and lung, but is also detected in most other tissues, except for the brain.
