Target description: | Serotransferrin (UniProt: P02787; also known as Transferrin, Beta-1 metal-binding globulin, Siderophilin) is encoded by the TF (also known as PRO1400) gene (Gene ID: in human. Transferrin is a monomeric glycoprotein that is expressed mainly in the liver and is secreted in plasma. Its functions are mediated via binding to transferrin receptor. It serves as an iron binding and transport protein that can bind two Fe3+ ions in association with the binding of an anion, usually a bicarbonate. It transports iron from sites of absorption and heme degradation to those of storage and utilization. Transferrin is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to produce the mature form. The mature form consists of alpha helices and beta sheets that form two domains. The N and C- terminal sequences are represented as globular N- and C- lobes that are connected with a short peptide. The carbohydrate moiety is attached to the C-lobe. Each lobe is further subdivided into two sub-domains- N1 and N2 for the N- lobe; and C1 and C2 for the C-lobe. Higher levels of transferrin are reported under conditions of anemia and during pregnancy. Mutations in TF gene are linked to Atransferrnemia that is characterized by abnormal synthesis of transferrin leading to iron overload and microcytic hypochromic anemia.
