Western Blotting Enhanced Validation - Recombinant Antibody Technology Untreated HeLa cell lysate (Lane 1) and HeLa cell lysate spiked with GST (Lane 2) were probed with Cat. No. ZRB1223, Anti-GST, clone 5B8 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates GST (~26 kDa).
Dot Blot Enhanced Validation - Recombinant Antibody Technology GST-Tagged recombinant protein (portion of CD25 from amino acids 22-140) and His-Tagged recombinant protein were probed with Cat. No. ZRB1223, Anti-GST, clone 5B8 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Lane 1: GST-Tagged Recombinant protein (portion of CD25 from amino acids 22-140 with a GST/His tag). Lane 2: His-Tagged Recombinant protein (portion of CD25 from amino acids 22-140 with only a His-tag).
ELISA Enhanced Validation - Recombinant Antibody Technology 0.5 μg/mL of GST, GST-TBP (Tata-binnding protein), GST-beta-catenin coated plates were analyzed by sandwich ELISA using Cat. No. ZRB1223, Anti-GST, clone 5B8 ZooMAb® Rabbit Monoclonal using a Goat Anti-Rabbit secondary antibody.

Application: Anti-GST, clone 5B8 ZooMAb, Cat. No. ZRB1223, is a recombinant Rabbit monoclonal antibody that specifically detects Glutathione S Transferase and has been tested for use in Dot Blot, ELISA, Immunocytochemistry and Western Blotting.
Application: Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected GST in HeLa cells transfeced with GST expression plasmid.

ELISA Analysis: 0.5 μg/mL from a representative lot detected GST on plates coated with in GST, GST-TBP (Tata-Bindidng protein), and GST-beta-catenin.

Dot Blot Analysis: A 1:1,000 dilution from a representative lot detected various concentrations of GST-Tagged recombinant protein (portion of CD25 from amino acids 22-140), but not the His-Tagged recombinant protein.

Note: Actual optimal working dilutions must be determined by the end user as specimens and experimental conditions may vary.

Target description: Glutathione S-transferase (also known as GST) is encoded by the Gst gene in E. coli. Glutathione S transferases are a group of enzymes that are responsible for the detoxification of reactive chemical species through conjugation to reduced glutathione. They are ubiquitously distributed in nature, being found in organisms from microbes to higher mammals. They exist as membrane-bound as well as cytosolic enzymes with distinct catalytic and noncatalytic binding properties. They catalyze the conjugation of reduced glutathione (GSH) with compounds that contain an electrophilic center through the formation of a thioether bond between the sulfur atom of GSH and the substrate. The fundamental basis of their catalytic activity has been attributed to their capacity to lower the pKa of the sulfhydryl group of GSH from 9.0 in aqueous solution to about 6.5 when GSH is bound in the active site. In the case of certain substrates, GST can catalyze both the forward and reverse reactions, which can lead to increased toxicity rather than detoxication. GST has also been used as a tag for recombinant proteins where it can be fused with either the N- or the C-terminal of protein. Proteins produced using the GST fusion system have been used in numerous biological applications. It allows proteins to become more soluble. Based on their biochemical, immunological, and structural properties, the soluble human GSTs are categorized into 4 main classes: alpha, mu, pi, and theta.
