Immunocytochemistry Enhanced Validation - Recombinant Antibody Technology Immunofluorescent analysis of Jurkat cells was performed using a 1:100 dilution of Cat. No. ZRB1199, Anti-Zfp64, clone 3B22 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488. Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.

Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from A549 cells (Lane 1) and Jurkat cells (Lane 2) were probed with Cat. No. ZRB1199, Anti-Zfp64, clone 3B22 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Zfp64 (~80 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or Zfp64 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1199, Anti-Zfp64, clone 3B22 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Zfp64 peptide displayed a KD of 1.1 x 10-8, whereas binding to non-specific control peptide displayed a KD of 6.8 x 10-4.

Target description: Zinc finger protein 64 (UniProt: Q9NPA5; also known as Zfp-64, Zinc finger protein 338) is encoded by the ZFP64 (also known as ZNF338) gene (Gene ID: 55734) in human. ZNF64 is a member of the krueppel C2H2 type zinc-finger protein family. It is a transcriptional regulatory protein with 13 zinc finger regions. ZFP64 has been identified as an essential transcription factor in MLL-rearranged leukemia where it maintains MLL expression via binding to the MLL promoter, which is the most enriched location of ZFP64. ZFP64 expression is reported to be under the control of Runx2, and it is upregulated by direct transactivation of its promoter. It is shown to associate with Notch 1 and suppress the myogenic differentiation of C2C12 cells and promotes their osteoblastic differentiation. ZFP64 is shown to promote Toll-like receptor-triggered proinflammatory and type I interferon production in macrophages by enhancing the activation of p65 subunit of NF-kB. ZFP64 can bind to the promoter of the TNF- , IL-6, and IFN-β genes in macrophages following TLR ligation. Upon stimulation of cells with lipopolysaccharide (LPS), ZFP64 associates with p65 subunit of NF-kB. Six different isoforms of ZFP64 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
