Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from PC12 (Lane 1) and NIH3T3 (Lane 2) cells were probed with Cat. No. ZRB1186, Anti-ZNF638, clone 4C19 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates ZNF638 (~220 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 μg/mL of non-specific control peptide (Run 1) or ZNF638 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1186, Anti-ZNF638, clone 4C19 ZooMAb® Rabbit Monoclonal. The binding of this antibody to ZNF638 peptide displayed a KD value of 4.1 x 10-7 whereas its binding to non-specific GST protein displayed a KD of 2.8 x 10-3.

Target description: Zinc finger protein 638 (UniProt: Q14966; also known as Cutaneous T-cell lymphoma-associated antigen se33-1, CTCL-associated antigen se33-1, Nuclear protein 220, Zinc finger matrin-like protein, ZNF638) is encoded by the ZNF638 (also known as NP220, ZFML) gene (Gene ID: 27332) in human. ZNF638 is a nucleocytoplasmic transcription factor that binds to cytidine clusters in double-stranded DNA and plays a key role in the silencing of unintegrated retroviral DNA. It is also shown to act as an early regulator of adipogenesis by acting as a transcription cofactor of CEBPs, controlling the expression of PPAR and other pro-adipogenic genes, such as SREBF1. ZNF638 is selectively expressed in mature thermogenic adipocytes and tissues and its induction in response to classic stimuli promotes heat generation. It is shown to be upregulated by cAMP modulators in vitro and by cold exposure and by pharmacological stimulation of β-adrenergic signaling in vivo. Ectopic expression of ZNF638 is reported to increase adipogenesis in vitro, whereas its knockdown inhibits differentiation and decreases the expression of adipocyte-specific genes. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
