Target description: | pro-ADM (UniProt: P35318; also known as Mid-regional pro-adrenomedullin) is encoded by the ADM (also known as AM) gene (Gene ID: 133) in human. ADM is synthesized with a signal peptide (aa 1-21) and is cleaved into Adrenomedullin (AM; aa 95-146) and Proadrenomedullin N-20 terminal peptide (PAMP; aa 21-41). AM and PAMP serve as potent hypotensive and vasodilatory agents involved in physiologic control of fluid and electrolyte homeostasis. In the kidney, AM acts as a diuretic and natriuretic, and both AM and PAMP are known to inhibit aldosterone secretion. ADM contains a region known as mid-region ProADM (aa 45-92) that lacks any activity but is a reliable biomarker for sepsis and progression to septic shock. ADM has been found in all epithelial surfaces that separate the internal and external environment, such as integument, respiratory, digestive and genito-urinary tract. It is also found in all bodily secretions and imparts protection against external pathogens. Its highest levels are reported in pheochromocytoma and adrenal medulla. ADM as well as PAMP also display potent antimicrobial action against both Gram-positive and Gram-negative bacteria. MR-proADM, a fragment of 48 amino acids that splits from the proADM and is present in a ratio of 1:1 with ADM, proportionally represents the levels and activity of ADM.
