Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Desmin (UniProt: P17661) is encoded by the DES (also known as CDCD3, CMD1F, CSM1) gene (Gene ID: 1674) in human. Desmin is a homopolymeric protein that is classified as a muscle-specific type III intermediate filament. It is shown to be essential for proper muscular structure and function. It plays a crucial role in maintaining the structure of sarcomeres, inter-connecting the Z-disks and forming the myofibrils, linking them not only to the sarcolemmal cytoskeleton, but also to the nucleus and mitochondria, thus providing strength for the muscle fiber during activity. It also acts as a sarcomeric microtubule-anchoring protein that specifically associates with detyrosinated tubulin-alpha chains, leading to buckled microtubules and mechanical resistance to contraction. Desmin can undergo phosphorylation at multiple serines by CDK1. Phosphorylation at serine 60 by Aurora B and threonine 76 by ROCK1 is reported to contribute to efficient separation of Desmin intermediate filaments during mitosis. Mutations in DES gene have been linked to myofibrillar myopathy that is characterized at ultrastructural level by disintegration of the sarcomeric Z disk and myofibrils, and replacement of the normal myofibrillar markings by small dense granules, or larger hyaline masses, or amorphous material. It leads to skeletal muscle weakness associated with cardiac conduction blocks, arrhythmias, and restrictive heart failure. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
