Immunofluorescence Enhanced Validation-Recombinant Antibody Technology Immunofluorescent analysis of human salivary gland tissue sections were performed using a 1:100 dilution of Cat. No. ZRB1171, Anti-TMEM16A, clone 3J18 ZooMAb® Rabbit Monoclonal and visualized with a secondary antibody conjugated to Alexa Fluor® 488 (Green). Nucleus is stained with DAPI (Blue). This antibody positively stains the cytoplasm.
Western Blotting Enhanced Validation-Recombinant Antibody Technology Human pancreas tissue lysate was probed with Cat. No. ZRB1171, Anti-TMEM16A, clone 3J18 ZooMAb® Rabbit Monoclonal (1:100 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates TMEM16A (~150 kDa).

Target description: Transmembrane protein 16A (UniProt: Q5XXA6; also known as TMEM16A, Anoctamin-1, Discovered on gastrointestinal stromal tumors protein 1, Oral cancer overexpressed protein 2, Tumor-amplified and overexpressed sequence 2) is encoded by the ANO1 (also known as DOG1, ORAOV2, TAOS2, TMEM16A) gene (Gene ID: 55107) in human. TMEM16A is a homodimeric multi-pass membrane protein that along with TMEMB16B subunit serves as the calcium-activated chloride channels (CaCC). It is broadly expressed with higher levels in liver, skeletal muscle, and gastrointestinal muscles. It plays a role in transepithelial anion transport and smooth muscle contraction and is also required for the normal functioning of the interstitial cells of Cajal (ICCs) that generate electrical pacemaker activity in gastrointestinal smooth muscles. It also acts as a major contributor to basal and stimulated chloride conductance in airway epithelial cells and plays an important role in tracheal cartilage development. ATP and calmodulin are shown to be essential for TMEM16A activation and the channel activity is inhibited by CFTR protein and by chloride channel inhibitors such as niflumic acid and 4, 4 -diisothiocyanatostilbene-2,2′-disulfonic acid (DIDS).
