Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from HeLa cells (Lane 1) and NIH3T3 cells (Lane 2) were probed with Cat. No. ZRB1161, Anti-ANKZF1, clone 1D4 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates ANKZF1 (~110 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or ANKZF1 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1161, Anti-ANKZF1, clone 1D4 ZooMAb® Rabbit Monoclonal. The binding of this antibody to ANKZF1 peptide displayed a KD value of 3.1 x 10-7, whereas its binding to non-specific peptide displayed a KD of 1.9 x 10-3.

Specificity: Clone 1D4 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Ankyrin repeat and zinc finger domain-containing protein 1 (ANKZF1). It targets an epitope within 18 amino acids from the N-terminal region.
Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
Target description: Ankyrin repeat and zinc finger domain-containing protein 1 (UniProt: Q9H8Y5; also known as Zinc finger protein 744, ZNF744) is encoded by the ANKZF1 (also known as ZNF744) gene (Gene ID: 55139) in human. ANKZF1 plays a role in the cellular response to hydrogen peroxide and in the maintenance of mitochondrial integrity under conditions of cellular stress. It contains two ANK repeats (aa 493-526 and 534-563). It is a diffusely distributed in the cytoplasm and translocates to the mitochondria upon cellular stress and its depletion is shown to reduce mitochondrial integrity and mitochondrial respiration under conditions of cellular stress. Loss of function mutations in ANKZF1 gene result in deregulation of mitochondrial integrity and may play a pathogenic role in the development of infantile-onset inflammatory bowel disease. Over-expression of ANKZF1 has been linked to poor overall survival in colon cancer subjects. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
