Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from human foreskin fibroblasts (Lane 1), C6 cells (Lane 2), and NIH/3T3 cells (Lane 3) were probed with Cat. No. ZRB1145, Anti-HSP60, clone 1N3 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates HSP60 (~61 kDa).

Target description: 60 kDa heat shock protein, mitochondrial (UniProt: P10809; also known as 60 kDa chaperonin, Chaperonin 60, CPN60, Heat shock protein 60, HSP-60, Hsp60, HuCHA60, Mitochondrial matrix protein P1, P60 lymphocyte protein) is encoded by the HSPD1 (also known as HSP60) gene (Gene ID: 3329) in human. Hsp60 is a homoheptameric, highly conserved molecular chaperone present in the mitochondrial matrix where along with Hsp10 it helps in protein folding. It also prevents misfolding of proteins and promotes refolding. It is transcribed by a nuclear gene, translated in the cytosol and the newly translated protein is imported into mitochondria. Its mitochondrial import signal (MIS) is localized in the 26 amino acids at the N-terminus. Following its import, the MIS peptide is cleaved off to generate the mature protein. Its mitochondrial import depends on the mitochondrial membrane potential. Under certain pathological conditions, such as cancer and chronic inflammatory diseases, Hsp60 can also accumulate in the cytosol via a mitochondrial export mechanism. Two isoforms of Hsp60 have been described that are produced by alternative splicing.
