Immunostaining was performed using a 1:1,000 dilution of Cat. No. ZRB1125, Anti-LC3A, clone 3J12 ZooMAb® Rabbit Monoclonal. Reactivity was detected using a Goat Anti-Rabbit IgG and HRP-DAB. Cytoplasmic/membranous staining was observed in neurons of human cerebral cortex sections.

Western Blotting Enhanced Validation - Recombinant Antibody Technology Lysates from Human Brain tissue (Lane 1), Mouse Brain tissue (Lane 2), Rat Brain tissue (Lane 3), and PC12 cells (Lane 4) lysates were probed with Cat. No. ZRB1125, Anti-LC3A, clone 3J12 ZooMAb® Rabbit Monoclonal (1:10,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates LC3A (~14 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or LC3A peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1125, Anti-LC3A, clone 3J12 ZooMAb® Rabbit Monoclonal. The binding of this antibody to LC3A peptide displayed a KD of 3.3 x 10-8 whereas binding to non-specific control peptide displayed a KD of 6.2 x 10-4.

Target description: Microtubule-associated proteins 1A/1B light chain 3A (UniProt: Q9H492; also known as Autophagy-related protein LC3A, Autophagy-related ubiquitin-like modifier LC3A, MAP1 light chain 3-like protein 1, MAP1A/MAP1B light chain 3 A, MAP1A/MAP1B LC3 A, Microtubule-associated protein 1 light chain 3 alpha) is encoded by the MAP1LC3A gene (Gene ID: 84557) in human. LC3s are structural proteins of autophagosomal membranes and are widely used as biomarkers of autophagy. They are ubiquitously distributed in all eukaryotes and are expressed as three splice variants known as LC3A, LC3B, and LC3C. LC3A is mainly localized to perinuclear and nuclear regions, LC3B is evenly distributed throughout the cytoplasm and is also localized in the nucleolar regions, and LC3C is shown to be located in the cytoplasm and strongly in the nuclei. Two variants (v1 and v2) of LC3A have been described that have different transcriptional start sites. Nuclear LC3A is shown to co-localize with Beclin-1. All three LC3s show a conserved region that is required for autophagosome formation. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
