Application: | Quality Control Testing Evaluated by Western Blotting in HeLa cell lysate. Western Blotting Analysis: A 1:1,000 dilution of this antibody detected QPRTase in HeLa cell lysate. Tested Applications Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected QPRTase in lysates from human brain, and C6, and HepG2 cells. Affinity Binding Assay: A representative lot of this antibody bound recombinant QPRTase with a KD of 1.3 x 10-7 in an affinity binding assay. Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected QPRTase in human kidney tissue sections. Flow Cytometry Analysis: 0.1 μg from a representative lot detected QPRTase in one million HepG2 cells. Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected QPRTase in HeLa cells. Target description: | Nicotinate-nucleotide pyrophosphorylase [carboxylating] (UniProt: Q15274; also known as EC:2.4.2.19, Quinolinate phosphoribosyltransferase [decarboxylating], QAPRTase, QPRTase) is encoded by the QPRT gene (Gene ID: 23475) in human. QPRTase is a hexameric enzyme formed by three homodimers. It is involved in de novo synthesis of NAD using quinolinic acid (QA) as a precursor that is produced via the degradation of tryptophan in kynurenine pathway. In mammals QA is shown to be an excitotoxic compound that can lead to neurodegeneration via over activation of NMDA receptors.
