Target description: CCAAT/enhancer-binding protein beta (UniProt: P17676; also known as C/EBP beta, Liver activator protein, LAP, Liver-enriched inhibitory protein, LIP, Nuclear factor NF-IL6, Transcription factor 5, TCF-5) is encoded by the CEBPB (also known as TCF5) gene (Gene ID: 1051) in human. C/EBP beta is a highly conserved B-Zip family member that acts as a transcription factor regulating the expression of genes involved in immune and inflammatory responses. It is expressed at low levels in the lung, kidney and spleen and is induced by endoplasmic reticulum stress. It can bind to DNA either as a homodimer or a heterodimer with ATF4. C/EBP beta plays a significant role in adipogenesis, as well as in the gluconeogenesis, liver regeneration, and hematopoiesis. During adipogenesis, it is rapidly expressed and, after activation by phosphorylation, induces CEBPA and PPARG, which turn on the series of adipocyte genes giving rise to the adipocyte phenotype. It has a pro-mitotic effect on many cell types, including hepatocytes and adipocytes, but has an anti-proliferative effect on T-cells by repressing MYC expression, facilitating differentiation along the T-helper 2 lineage. C/EBP beta is phosphorylated at threonine 235 by MAPK and CDK2 that serves to prime phosphorylation at threonine 226 and serine 231 by GSK3beta and acquire DNA-binding as well as transactivation activities that are required to induce adipogenesis. However, O-glycosylation at serine 227 and serine 228 is shown to prevent phosphorylation on threonine 235, serine 231, and threonine 226 and reduces its DNA binding activity that delays adipocyte differentiation.
