Immunostaining was performed using a 1:10,000 dilution of Cat. No. ZRB1098, Anti-Brn3a, clone 4L5 ZooMAb® Rabbit Monoclonal. Reactivity was detected using an Goat Anti-Rabbit and HRP-DAB. In human retina, nuclear staining was observed in subsets of cells within GCL & ONL, while membranous staining was observed in areas extending from IPL, INL & OPL (Middle image). In mouse retina, nuclear staining was observed mostly in INL, with a lesser degree of staining in GCL (B).

Western Blotting Enhanced Validation-Recombinant Antibody Technology Human (Lane 1), mouse (Lane 2), and rat (Lane 3) brain tissue lysates were probed with Cat. No. ZRB1098, Anti-Brn3a, clone 4L5 ZooMAb® Rabbit Monoclonal (1:1,000 (Lane 1) and 1:10,000 (Lane 2 and 2)). Proteins were visualized using a Donkey Anti-Rabbit IgG + secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Brn3a (~48 kDa).

Target description: POU domain, class 4, transcription factor 1 (UniProt: Q01851; also known as Brain-specific homeobox/POU domain protein 3A, Brain-3A, Brn-3A, Homeobox/POU domain protein RDC-1, Oct-T1) is encoded by the POU4F1 (also known as BRN3A, RDC1) gene (Gene ID: 5457) in human. Brn-3A is a multifunctional transcription factor with different regions mediating its different effects. It regulates the expression of specific genes involved in differentiation and survival within a subset of neuronal lineages. It is expressed in the brain and the retina and is present in the developing brain, spinal cord and eye. Its expression peaks early in embryogenesis and is undetectable 14 days after birth. Its C-terminal domain can act as both DNA-binding domain and a transcriptional activator. The N-terminal domain is also required for transactivation activity on some target genes acting as a discrete activation domain. Brn-3A acts by binding to sequences related to the consensus octamer motif 5′-ATGCAAAT-3′ in the regulatory regions of its target genes. The POU-specific domain of Brn-3A is localized to amino acids 261-338. Brn-3A is shown to activate Bcl-2 expression and protect neuronal cells from apoptosis. Two isoforms of Brn-3A have been described that are produced by alternative splicing. Isoform 2 can act as a transcription factor but cannot regulate the expression of the same subset of genes and does not have any anti-apoptotic effect on neuronal cells. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
