Western Blotting Enhanced Validation - Recombinant Antibody Technology Nuclear extracts from HEK293 cells (Lane 1), NIH3T3 cells (Lane 2), HeLa cells (Lane 3), and L6 (Lane 4) were probed with Cat. No. ZRB1084, Anti-CBX2, clone 3I14 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates CBX2 (~56 kDa).
Affinity Binding Assay Enhanced Validation - Recombinant Antibody Technology 100 mg/mL of non-specific control peptide (Run 1) or CBX2 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1084, Anti-CBX2, clone 3I14 ZooMAb® Rabbit Monoclonal. The binding of this antibody to CBX2 peptide displayed a KD of 8.7 x 10-7, whereas binding to non-specific control peptide displayed a KD of 2.9 x 10-3.

Target description: Chromobox protein homolog 2 (UniProt: Q14781; also known as CBX2) is encoded by the CBX gene (Gene ID: 84733) in human. CBX2 is a component of a Polycomb group (PcG) multiprotein PRC1-like complex that is expressed during interphase and metaphase. It plays a role in the lineage differentiation of the germ layers in embryonic development and is involved in sexual development, acting as activator of NR5A1 expression. It is shown to be localized in distinct foci on chromatin and in chromocenters. CBX2 is reported to be expressed from week 9 to week 25 post-fertilization in both male and female gonads. It is required to maintain the transcriptionally repressive state of many genes throughout development via chromatin remodeling and modification of histones. Disruption of this gene in murine models is shown to result in male-to-female gonadal sex reversal. Levels of CBX2 are reported to decrease in macrophages upon viral infection and knockdown of CBX2 gene in macrophages can inhibit virus-induced production of interferon-. CBX2 is reported to bind and recruit Lysine demethylase 6B (Jmjd3) to the IFN- promoter, leading to demethylation of H3K27me3 and increased transcription of IFN-β. Mutations in CBX2 gene are associated with gonadal dysgenesis in humans. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
