Immunocytochemistry Enhanced Validation-Recombinant Antibody Technology Immunofluorescent analysis of MCF-7 cells was performed using a 1:100 dilution of Cat. No. ZRB1071, Anti-Ubiquilin 4, clone 5K6 ZooMAb® Rabbit Monoclonal and visualized with a Goat Anti-Rabbit secondary antibody conjugated to Alexa Fluor® 488. Actin filaments have been labeled with phalloidin (Red). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.

Western Blotting Enhanced Validation-Recombinant Antibody Technology Raji nuclear extract (Lane 1), HeLa nuclear extract (Lane 2), and HepG2 nuclear extract (Lane 3) were probed with Cat. No. ZRB1071, Anti-Ubiquilin 4, clone 5K6 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Ubiquilin 4 (~64 kDa).

Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of Ubiquilin 4 peptide (Run 1) and non-specific control peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1071, Anti-Ubiquilin 4, clone 5K6 ZooMAb® Rabbit Monoclonal. The binding of this antibody to Ubiquilin 4 peptide displayed a KD value of 4.0 x 10-7 whereas its binding to non-specific control peptide displayed a KD of 5.7 x 10-3.

Target description:	Ubiquilin-4 (UniProt: Q9NRR5; also known as Ataxin-1 interacting ubiquitin-like protein, A1Up, Ataxin-1 ubiquitin-like-interacting protein, A1U, Connexin43-interacting protein of 75 kDa, CIP75) is encoded by the UBQLN4 (also known as C1orf6, CIP75, UBIN) gene (Gene ID: 56893) in human. Ubiquilin-4, a member of the UbL-UBA protein family, serves as an adaptor in the degradation of specific substrates via the proteasomal pathway. It is highly expressed in pancreas, kidney, skeletal muscle, heart, and throughout the brain. Its levels are reported to be upregulated in aggressive tumors. It serves as a regulator of protein degradation and mediates the proteasomal targeting of misfolded, mislocalized or accumulated proteins. It acts by binding polyubiquitin chains of target proteins via its UBA domain and by interacting with subunits of the proteasome via its ubiquitin-like domain. Ubiquilin-4 colocalizes with the proteasome, both in nucleus and cytoplasm and is exported from the nucleus following interaction with DESI1/POST. In response to DNA damage and phosphorylation at serine 318 by ATM, it localizes to the nucleus and is recruited to sites of DNA damage where it represses homologous recombination repair. It acts by binding and removing ubiquitinated MRE11 from damaged chromatin, leading to MRE11 degradation by the proteasome. MRE11 degradation then prevents homologous recombination repair and redirects double-strand break repair toward non-homologous end joining. Mutations in UBQLN4 gene are shown to cause Amyotrophic lateral sclerosis (ALS). Defects in UBQLN4 gene also cause intellectual impairment, growth retardation, microcephaly, and facial dysmorphism.
