Western Blotting Enhanced Validation-Recombinant Antibody Technology Lysates from HeLa (Lane 1), NIH3T3 (Lane 2), Jurkat (Lane 3), and L6 (Lane 4) cells were probed with Cat. No. ZRB1068, Anti-PRPF19, clone 1K10 ZooMAb® Rabbit Monoclonal (1:1,000 dilution). Proteins were visualized using a Donkey Anti-Rabbit IgG secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates PRPF19 (~55 kDa).
Affinity Binding Assay Enhanced Validation-Recombinant Antibody Technology 100 μg/mL of control peptide (Run 1) or PRPF19 peptide (Run 2) were analyzed by affinity binding assay using Cat. No. ZRB1068, Anti-PRPF19, clone 1K10 ZooMAb® Rabbit Monoclonal. The binding of this antibody to PRPF19 peptide displayed a KD value of 2.4 x 10-6 whereas its binding to control peptide displayed a KD of 1.0 x 10-2.

Target description: Pre-mRNA-processing factor 19 (UniProt: Q9UMS4; also known as EC:2.3.2.27, Nuclear matrix protein 200, PRP19/PSO4 homolog, hPso4, RING-type E3 ubiquitin transferase PRP19, Senescence evasion factor, PRPF19) is encoded by the PRPF19 (also known as NMP200, PRP19, SNEV) gene (Gene ID: 27339) in human. PRPF19 is a homotetrameric protein that is ubiquitously expressed. Its expression in shown to be elevated in tumor cell lines. PRPF19 is a ubiquitin-protein ligase which is a core component of several complexes mainly involved pre-mRNA splicing and DNA repair. It is required for pre-mRNA splicing as component of the spliceosome. During assembly of the spliceosome it mediates lysine 63-linked polyubiquitination of the U4 spliceosomal protein PRPF3 and ubiquitination of PRPF3 allows its recognition by the U5 component PRPF8 and stabilizes the U4/U5/U6 tri-snRNP spliceosomal complex. PRPF19 distribution varies with the cell cycle stage. It is shown to be nucleoplasmic in interphase cells and is irregularly distributed in anaphase cells. In the prophase stage it is uniformly distributed and in metaphase stage it is found in the extrachromosomal regions.
